How to apply boundary condition in finite element

As part of the finite element problem with a two-node element, I want to construct the boundary vector:
fsur = [10*q(x=2); -150];
...where q(x=2) is the unknown heat flux in node 1. How do you construct this vector, since q is an undefined and unknown variable.
