Film thickness and a change in film thickness measurements for a number of images taken at different times at the same camera position? Thickness should be in metric units

Hi fellows,

I am intended to measure the change in a film or deposit thikcness change over time using high speed, high resultion camera (i.e. online measurement of thickness cahnge). During the run of the experiment, a number of images will be taken at different time intervals for the depostion surfcace, where the film thickness is expected to reduce. I am wondering how I would do that using MATLAB?

Are segmentation and edge detection enough? How do I calibrate the image to get actual values of the measurements in metric units?
