I don't see any colored 5x5 or 9x9 regions there. But I do remember seeing that picture sometime recently. Like I told renuka you can just use convolution to find out the sums everywhere, or if you have known regions, like the red, you can just use sum() and indexing:

subImage = yourImage(4:6,1:3);
theSum = sum(subImage(:));
