given when extracting ROI of mouth after enhancement into logical image, it become imperfect mouth shape, which the mouth is separated into two. how do i want to get better image which show a complete mouth shape?

Who knows (not having seen your image)?
Maybe you can use activecontour(), demo attached.
Maybe you can find the centroid and just plop down a template there, replacing what you've got with a "perfect" version of it from a library of perfect shapes.
I don't know - I'm not as good working "blind".
