My GUI application needs to access a database (about 50 MB, stored as a .mat file). Currently, I store the data in the gui handles and use 'guidata' to update the data. However, it greatly slows down the speed of the GUI. Any solution to this problems? Thanks a lot!

50 MB is not a large array in times of Gigabytes of RAM. Sharing an array by guidata does not create deep copies, but Matlab uses an efficient Copy-on-Write method. In consequence it is not clear, what exactly slows down your code. Use the profiler to find the bottleneck and post a minimal working example, which reproduces the problem.
