- What is winspec (your first program)?
- In what 2nd program are you opening it and finding it's darker? Are you basing this off the appearance of the image once displayed in that second program, or by inspecting the digital values?
- Then, after that second program you say you consequently open it in MATLAB (the 3rd program) and find it's darker than the second program. Again, are you basing this off the appearance of the image once displayed in MATLAB, or by inspecting the digital values in the workspace?
- Can you attach your .tiff image and tell us for the upper left (1,1) pixel, what winspec says the value is, what the second program says the value is, and what MATLAB says the value is?

After I save .tiff file from winspec and if I open it the image gets darker and consequently if i read them through matlab, I get very less intensity values. I am not getting the correct intensity values since it is dark. It is a 16 bit image .
