Help: how to count the number of holes on the images and its sizes

I attached a file containing image.. It consists of many black holes.. I want to count the number of black holes and its sizes.. I am very new to the image processing..It will be helpful it you provide some code in executing this.. Any help is much appreciated..

The input image seems a bit challenging, yet you can try the following basic methods and check the results.

One way:

1. Convert the image to binary with best fit thresholding (or see other segmentation appoaches)
2. Use bwlabel

[L,n] = bwlabel(binary_image) % here n gives the number of connected objects 

or

2. For measuring the properties of retions, like calculae area, you can look at regionprops
